Abstract

Fungi have an extraordinary ability to degrade a broad range of toxic chemicals via oxidative and reductive pathways. Limited literature is available on the cytochrome p450 (CYP) dependent monooxygenase system of filamentous fungi. In this paper, we studied the basal expression of CYP and its inducibility by various carbon sources, including some n-alkanes and fatty acids, in Trichoderma harzianum. We observed that n-dodecane and a mixture of fatty acids were good inducers of total CYP content in T. harzianum and isolated by RACE an open reading frame of 1520 bp which encoded a CYP52 protein of 520 amino acids. A further inducible CYP was identified in a library of T. harzianum via the peptides found by LC-MS/Ms analysis of a protein band induced by n-dodecane exposure. Thus, the filamentous fungus T. harzianum could be expected to convert alkanes to fatty acids and to incorporate them into cellular lipids.
