Abstract
(English)
A time-dependent loss of ribulose-1,5-bisphosphate carboxylase (RuBPcase) activity and partial proteolysis were observed in squeezed juices of Jerusalem artichoke (Helianthus tuberosusL.) obtained by the typical wet green crop fractionation procedure. The use of protease inhibitors resulted in diminished proteolysis with no effect on the decay of enzyme activity. The RuBPcase activity was preserved by addition of thiol reagents to the squeezed juices. The relationship between the number of oxidized thiol groups and residual activity was investigated in purified preparations of RuBPcase and the functional role of thiol groups for the maintenance of the native structure of RuBPcase is discussed.
